As you are preparing to send your manuscript off to literary agencies and publishing houses, your query letter is the first impression and sometimes your only shot to get your book read. If your query letter generates enough interest and intrigue, the agent or publisher will request to read the first three chapters or the full manuscript. Having interned at a literary agency, I have had experience reading query letters and know what are the most important things to highlight. Every place you submit to will have it's own set of rules and it is important to follow those rules and cater each query letter to that particular company.
